The third exon was replaced with an enhanced GFP gene. Quantitative real-time PCR confirmed absence of Prnp mRNA in homozygotes and lower levels, about 50-60% that of wild-type, in heterozygotes. GFP expression was confirmed in various regions of the brain and in astrocytes. (J:215179)
