This mutation was created by generating targeting constructs containing a 5' homology region with Pten exons 4-5, a neo cassette flanked by two loxP sites (LoxP- Neo-LoxP), and a 3' homology region containing exon 6 with the point mutation (c.565A > T) preceded by a FLAG tag. After homologous recombination in ES cells and injection into C57BL/6 blastocysts, the resulting chimeras were bred with EIIA-Cre transgenic female mice to delete the loxP-flanked neomycin resistance gene (neo) from the targeted allele. The C-terminal truncated Pten protein, is expressed in various tissues including kidney, lung, uterus and heart. (J:211716)
