The bacterial artificial chromosome RP23-431D9, which contains the entire mouse choline acetyltransferase gene, was modified by insertion at the initiating ATG codon, in exon 3, of a DNA sequence encoding a tau/enhanced green fluorescent protein (tau/EGFP) fusion protein followed by a triple polyadenylation signal and a kanamycin resistance gene (aphAI). Detailed comparison of EGFP fluorescence/immunohistochemical staining and endogenous CHAT expression, visualized immunohistochemically, demonstrates their almost complete co-localization throughout the brain and peripheral nervous system; as anticipated, fusion to tau localizes EGFP to axons.
