A loxP site and a neo-tk cassette flanked by loxP sites were inserted into the MscI site of the 5' untranslated region and NheI site within the first intron, respectively. The loxP flanked neo-tk cassette was removed via Cre-mediated recombination, leaving the first coding exon flanked by loxP sites. (J:214945)
