A loxP flanked PGK-Neo cassette was inserted into the intron between exons 1b and 2, a cre-expressing plasmid was introduced to delete the neomycin cassette, leaving a single loxP site behind, and a loxP-ires-tau:Egfp; FRT-PGK-Neo-FRT cassette was inserted immediately after the stop codon in the 3'UTR of exon 13.
