A lox66 was inserted upstream of exon 2. An inverted STOP element and lox71 site was inserted downstream of exon 2. Flp-mediated recombination removed the FRT-flanked neomycin resistance cassette inserted between exon 2 and 3. Cre-mediated recombination restores the orientation of the STOP element and inverts exon 2. (J:209284)
