The transgene contains the promoter of the mouse myosin, heavy polypeptide 6, cardiac muscle, alpha (Myh6) gene joined to a cDNA encompassing the entire coding sequence of human adenylate cyclase 8 - which normally is expressed in the nervous system and is activated by Ca+ - followed by the SV40 intron and polyadenylation signal. Two transgenic founder lines were identified; both lines were estimated by dot-blot analysis to carry 7 to 8 copies of the transgene. Northern blot analysis of total RNA extracted from brain, heart, kidney and skeletal muscle of transgenic mice confirmed expression only in heart. Adenylate cyclase activity is increased at least 7-fold in cardiac membranes of transgenic vs. nontransgenic control mice. (J:62373)
