The mitoDendra fusion protein is composed of a cleavable N-terminal mitochondrial targeting pre-sequence (derived from human cytochrome c oxidase subunit VIII [COX8A]), a linker peptide (to improve fusion protein cleavage) and the monomeric photo-activateable fluorescent protein Dendra (derived from octocoral Dendronephthya sp.). The mitoDendra fusion protein cDNA sequence was placed downstream of the modified regulatory region of the "murine thy1.2 gene" (neuronal-specific Thy1.2 promoter; extending from the promoter to the intron following exon 4, excluding exon 3 and its flanking introns).
