One loxP site was inserted in intron 3 while a neo resistance cassette flanked with both loxP and an FRT sites (in that order) was inserted in intron 4. The resistance cassette was subsequently removed by expression of flp recombinase leaving exon 4 flanked by loxP sites. (J:206152)
