An FRT flanked lacZ/neo construct along with a loxP site was inserted upstream of exon 5 and a second loxP site was left in place 3' of exon 5. The 5' selection cassette was then removed by flp recombinase expression leaving exon 5 flanked by loxP sites. Subsequent cre expression excised exon 5. (J:206155)
