A codon-optimized cDNA encoding the salvage enzyme uracil phosphoribosyltransferase (UPRT) from the protozoan parasite Toxoplasma gondii is expressed under the control of the Gt(ROSA)26Sor promoter upon activation by cre recombinase. In cells expressing cre recombinase, and thus T gondii UPRT, 4-thiouracil injected into the mouse is converted to 4-thiouridine that is then incorporated into mRNA; this facilitates isolation and characterization of the transcriptome of a specifc cell population.
