Transgenic mice were produced by coinjecting embryos with the rearranged immunoglobulin kappa light chain gene of the hen egg lysozyme (HEL) binding hybridoma, HyHEL10, along with a chimeric IgM/IgG1 immunoglobulin heavy chain gene whose VDJH variable region elements also were derived from HyHEL10. The constant region of the expressed chimeric heavy chain comprises the CH1 and Fc regions from IgM and the extracellular spacer, transmembrane and and cytoplasmic domains of IgG1. Two copies of each transgene are estimated to be present in mice of this line based on densitometric analysis of Southern blots of tail DNA sequentially hybridized with probes from the germline Igh-J4 and Igk-J4 regions. Flow cytometric analysis of spleen cells incubated with labeled HEL demonstrated high-affinity HEL binding of ~48 +/- 27% of splenic B cells of this line. (J:61118)
