A BAC (RP24-166N8) containing the Ltf gene was modified in bacterial cells to produce the targeting construct. A cassette containing codon-improved cre recombinase (icre), an artificial intron, the bovine growth hormone polyadenylation signal and an frt-flanked ampicillin selection marker was inserted into the first exon of Ltf. A plasmid endoding Flp was transfected into E. coli cells to delete the amp-marker. The BAC construct was verified by sequencing and microinjected into pronuclei of (B6/129Sv)F1 oocytes to produce transgenic animals. 2 lines (14 and 15) were obtained.
