The full-length human huntingtin cDNA containing 48 CAG repeats is under the control of the cytomegalovirus promoter and SV40 enhancer. Three lines, B-D, were established. No CAG repeat instability was detected by PCR amplification. Western blot analysis confirmed transgene expression in the brain, including primary and secondary motor cortices, entorhinal cortex, striatum, hippocampus and cerebellum. Transgene expression in the brain is five times that of endogenous levels. (J:50173)
