Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a BAC (#RP23-309G21) containing the entire mouse Cspg4 (chondroitin sulfate proteoglycan 4) gene. The BAC was modified to create a membrane anchored form of EGFP (enhanced green fluorescent protein) under the control of the Cspg4 promoter followed by a polyadenylation sequence. EGFP is targeted to the plasma membrane by fusing the first 26 amino acids (containing two palmitoylation domains and a myristoylation domain) of Lck (lymphocyte-specific protein tyrosine kinase) to the EGFP N terminal.
