A single targeting event replaced most of exon 1 of Psmb9 with an in frame alkaline phosphatase floxed neo construct and exons 1 - 5 of Psmb8 with a lacZ poly A FRT construct. Immunoblot analysis confirmed the lack of expression of both PSMB9 and PSMB8 in lysates or proteasome preparations from spleens of homozygous mice. In addition expression of Tap1, located between the 2 targeted genes, mRNA and protein were reduced by about 50% in homozygous mice. (J:180765)
