A MICER clone carrying exons 1 and 2 of the HPRT gene, a neo cassette, a loxP site and Tyrosinase gene was inserted downstream of Defb13 and a MICER clone carrying exons 3-9 of the HPRT gene, a puro cassette, a loxP site and Agouti gene was inserted upstream of Defb1. Cre mediated recombination removed a 175 kb region encompassing 9 beta defensin genes (Defb1, Defb50, Defb2, Defb10, Defb9, Defb11, Defb15, Defb35 and Defb13) and reconstituting an intact Hprt gene. RT-PCR analysis confirmed the absence of expression of the nine deleted genes in homozygous mice. (J:204187)
