The cassette used to generate the targeting vector contained the CAG promoter, the EGFP sequence with loxP sites flanking the coding region, a rabbit polyA signal, and a FLAG-tagged tandem version of the DsRed fluorophore (tDsRed) with the SV40 polyA signal. This construct, ligated to homology arms and an Frt-flanked PGK-neo selection marker, was linearized and electroporated into ES cells for homologous recombination with the Gt(ROSA)26Sor locus.
