A targeting vector was designed to insert an FRT-flanked neomycin marker and a loxP site upstream of exon 2 of the Gfi1b gene and to insert a second loxP site between exons 5 and 6. Correctly targeted ES cells were treated with Flp to excise the neo resistance marker. (J:207446)
