A loxP site was inserted upstream of a modified exon 2 in which nucleotide substitutions (AAG to GCG) results in the amino acid substitution of alanine for lysine at position 127 (K127A). Flp-mediated recombination removed the FRT- and loxP-flanked neomycin resistance cassette inserted downstream of the modified exon 2. The product of this allele is predicted to be a kinase dead form.
