A recombinase mediated exchange (RMCE) vector was designed to contain (from 5' to 3') a promoterless neomycin cassette followed by 3 SV40 polyadenylation sites (pA), 3xpA, and in reverse orientation a fragment containing 3xIns (400 bp core region of chicken beta-globin insulator), CAGGS (CMV-IE enhancer/chicken beta-actin/rabbit beta-globin) hybrid promoter, floxed-3xpA, and a cDNA encoding an enhanced green fluorescent protein (EGFP) fused to ribosomal protein unit L10a (Rpl10a). The loxP-flanked pA cassette is designed to block transcription of EGFP/Rpl10a. (J:206130)
