A cDNA containing the tethered homodimer sequence followed by a flexible linker was inserted into exon 1 downstream of the start site methionine. Cre-mediated recombination removed the floxed puromycin resistance cassette inserted downstream of exon 1. Western blot analysis confirmed the absence of monomer protein in E10.5 or E8.5 embryos.
