BAC clone RP24-289D15 was modified by ET recombineering to insert a cassette with a hybrid intron, cre recombinase, an IRES site and the beta-galactosidase sequence (lacZ), along with an FRT-flanked neomycin/kanamycin selection marker, into the ATG site of the first coding exon of pre-proenkephalin (Penk). The selection marker was removed using Flp recombinase. The modified circular BAC was purified and verified by sequencing, before injection into oocytes. Founder information is not available.
