A targeting vector was constructed with a floxed transcriptional stop sequence, the Trp53 coding exons except for exon AS (Alternative Splicing), and a diphtheria toxin (DTA) gene. The linearized vector was electroporated into ES cells to undergo homologous recombination. Germline transmission of the mutated coding sequence was verified. (J:199308)
