A targeting vector was generated with a nonsense mutation in exon 10 that would delete the final 31 amino acids of the Trp53 gene. Upstream of the coding sequence was a transcriptional stop sequence and neo resistance marker flanked by loxP sites. The linearized targeting construct was electroporated into ES cells for homologous recombination. Chimeras were produced from injected blastocysts and germline transmission was verified. (J:199308)
