A floxed neo/Tk selection cassette was inserted upstream of exon 7 and a third loxP site was inserted downstream. The selection cassette and exon 7 were then excised from properly targeted ES cells by transient cre expression. Protein and mRNA were not detected in islets of knockout mice. (J:197386)
