This transgene consists of the full-length mouse Tcf4 open reading frame under the control of the Thy1.2 minigene, which restricts expression to the postnatal brain. In addition a Flag-tag was added on the N-terminus and a TAP-tag on the C-terminus of Tcf4. qRT-PCR shows approximately 1.5-fold overexpression in the hippocampus and expression in the cortex and amygdala but not in the striatum. Western blotting confirmed protein expression in the cortex and hippocampus but not in the cerebellum and in situ hybridization shows expression in the hippocampus, the neocortex, thalamus, and mesencephalic structures of adults.
