An FRT flanked neo cassette followed by a loxP site was inserted into intron 2. A second loxP site was inserted in intron 1. ES cells targeted to th NOD chromosome were selected. The neo selection cassette was then removed from properly targeted ES cells by flp recombinase expression leaving exon 2 flanked by loxP sites. (J:196160)
