Exon 3, which encodes the transmembrane domain, was replaced by the in-frame insertion of the EGFP coding sequence followed by a neomycin selection cassette in reverse orientation. The resulting EGFP fusion protein was unstable. No mRNA was detected in homozygous mice. (J:195536)
