A targeting vector was obtained from the European knockout consortium (EUCOMM). The targeting construct contains a splice acceptor, beta-geo cassette flanked by two Flp sites preceding the third exon. In addition, the targeting construct introduced two loxP sites on either side of exon 3. Transient cre expression resulted in the excision of exon 3 while leaving the FRT flanked beta-geo intact.
