A targeting construct inserted in the 5' UTR in exon 1. The vector contains a loxP site, HA-tagged cDNA with (a) nucleotide substitution(s) (and complete with TGA stop codon and polyadenylation sequence), FRT-flanked neomycin resistance cassette and loxP site. The point mutation results in the amino acid substitution of isoleucine for phenylalanine at position 77 (F77I). The predicted protein product is zolpidem-insensitive. Flp-mediated recombination removed the neomycin resistance cassette. In situ hybridization confirmed reduction of transcript expression in the brain. (J:199326)
