A targeting vector introduced the beta galactosidase coding sequence and an frt-flanked PGK-neo selection cassette into the Shox2 locus via homologous recombination in ES cells, replacing exon 1, intron 1 and part of exon 2. Germline animals were crossed to Flp deleter mice to remove the PGK-neo cassette. (J:199319)
