A floxed PGK-neo cassette was inserted 417 bp downstream of the gammaC1 exon and an additional loxP site was inserted 1026 bp upstream. The selection cassette and gammaC1 exon were removed by cre excision from properly targeted mice. In addition, this created a frame shift mutation. mRNA production was severely impaired in heterozygotes. (J:98737)
