A BAC clone (RP23-34C7) containing the Peo1 (Twinkle) sequence was used to generate the transgenic construct. Recombineering methods introduced a synonynmous change in the coding sequence which created an XhoI restriction site in exon 1. The purified construct was microinjected into pronuclei of one-stage mouse embryos. Line information is not available.
