The targeting construct used has lox recombination sites flanking the splice acceptor of a beta-geo fusion gene that was inserted into intron 1. In the unrecombined state, the presence of the beta-geo fusion gene disrupts expression of the gene. Upon recombination, splicing and expression of the endogenous gene can be restored. (J:196476)
