A cDNA encoding Aco1 (Irp1) was mutated by PCR so that 3 cysteines (C437, C503 and C506) were changed to serines and another cysteine (C118) to alanine. These mutations putatively stabilize the apoprotein in the presence of heme. The mutated cDNA (Aco1*) was fused to a FLAG tag and inserted into a plasmid behind a loxP-flanked STOP sequence. The final construct was electroporated into E14 ES cells for homologous recombination. Correctly targeted ES cells were used to generate mice carrying the conditional allele. These animals were crossed to Hprttm1(cre)Mnn mice to delete the STOP cassette. (J:196374)
