The endogenous coding sequence was replaced with a mutated Or10j5 sequence, in which the DRY motif was replaced by RDY (D121R;R122D), followed by an internal ribosome entry site and a constitutively active form of Gnas, followed by another internal ribosome entry site and a Tau-tdTomato reporter, in addition to a self-excising neomycin selection cassette (ACNF), thus encoding a tri-cistronic message that has a signaling-deficient OR10J5 olfactory receptor, a constitutively active GNAS to rescue G-protein signaling, and a fluorescent reporter.
