A loxP site was inserted upstream of exon 3 and a floxed pgk-hygromycin cassette was inserted downstream of exon 3 via homologous recombination. Cre mediated recombination removed exon 3 and the pgk-hygromycin cassette. Quantitative PCR and western blot analysis indicate the absence of mRNA and protein expression in the inner ear of homozygous mice. Expression of Gjb2 mRNA and protein are reduced in the inner ear of homozygous mice but to a lesser extent than in mice homozygous for Gjb6tm1Kwi. (J:193913)
