Sequence encoding for 6 histidines, a trombine cleavage site and a hemagglutinin (HA) tag was inserted in exon 1 in frame 3' of the start codon and a floxed neo cassette was inserted in intron 1 via homologous recombination. Cre mediated recombination removed the neo cassette. A fusion protein is detected in heterozygous mice. Presence of the tag does not appear to effect substrate specificity of SUMO1 conjugation.
