A cassette containing a loxP site followed by an FRT flanked neomycin resistance gene was inserted 288 bp upstream of exon2 while a second loxP site was inserted 97 bp downstream of exon 2. The neomycin genes was subsequently removed from correctly targeted ES cells using an adenovirus expressing flp recombinase. Exon2 was left flanked by loxP sites. (J:193283)
