A cassette containing an IRES-cre cDNA sequence and frt-flanked PGK-hygromycin selection marker was inserted into the 3' UTR (after the stop codon) of the Nfatc1 gene in R1 ES cells. Correctly targeted cells were selected and expanded, followed by blastocyst injection. Two lines were selected and crossed to E2A-flpe expressing mice to remove the selection marker. (J:193198)
