A tetO sequence followed by a loxP site and then an FRT flanked neomycin selection cassette were inserted immediately upstream of the Cd4 silencer sequence in intron 1. A second loxP site was inserted immediately downstream of the silencer. The selection cassette was subsequently removed by flp recombinase expression. (J:191711)
