A construct was inserted that uses the human ubiquitin C promoter to potentially drive expression of one of two variant forms of hen-egg lysozyme (HEL). A floxed polyadenylation sequence separates the promoter from the first FRT flanked HEL3x sequence and another polyadenylation sequence. This was followed by the HEL4x sequence and a third polyadenylation sequence. HEL4x differs from HEL3x by the presence of an additional K97R mutation. Cre expression places HEL3x under the control of the human UBC promoter. Flp expression followed by cre expression removes the HEL3x sequence and places HEL4x under the control of the promoter.
