A 1740 bp region that lies 335 kb upstream of the transcription start site of Myc (Chr15:61449842-61451581 (NCBI37/mm9)) was flanked with loxP sites and an FRT site flanked neomycin resistance gene cassette was inserted upstream of the 3' loxP site. Cre-mediated recombination removed the 1740 bp region and the neo cassette, leaving a single loxP site at chr15:61490136 (GRCm39). Expression of Myc RNA was decreased in the colon but not duodenum of homozygous mice at P1. Chromatin immunoprecipitation analysis indicated the loss of TCF7L2 binding in this region. (J:191217)
