alpha-Tpm1 cDNA was subjected to site-directed mutagenesis to generate a GAC to AAC transition at codon 175, which is predicted to result in an Asp to Asn amino acid change in the encoded protein. The mutated cDNA was then inserted downstream of the alpha-MHC promoter, which confers cardiac-specific expression. Transgenic lines 69, 108, 109, and 113, were produced and were selected for additional studies. (J:57365)
