The ES cells were targeted in the 3' UTR of the Col1a1 locus with a Flp-in construct containing a frt flanked PGK-Neo cassette followed by a Hygromicin gene lacking a proper ATG. Correctly targeted ES cells were selected on basis of Neomycin resistance. The targeted Col1a1 locus can later be used as a docking site for the introduction of a plasmid, minimally containing the desired transgene sequence and an frt site preceded by a constitutive promoter and an ATG initiation codon.
