By homologous recombination in ES cells, the targeting vector inserted a floxed neomycin (neo) resistance cassette into the 4th intron of the Relb gene and a single loxP site into 184 bp upstream of exon 4. The neo cassette was deleted by transient transfection of a plasmid (pMC-cre) expressing Cre recombinase in the ES cells, resulting in the allele with the exon 4 being floxed.
