Using reccombinase mediated cassette exchange (RMCE) using Pdx1tm1Mgn, a FRT-flanked hygro cassette was inserted upstream of Area IV and nuclear localized CFP was inserted into exon 1 at the translation initiation site replacing part of the exon. Flp-mediated recombination removed the hygro cassette. A lox66/71 site remains upstream of Area IV and a lox2272 site downstream of CFP. (J:187872)
