Cre recombinase was inserted into intron 14 of the Etl4 (Skt) locus by recombinase mediated cassette exchange using an exchangeable gene trap system using left element/right element (LE/RE) mutant lox sites. The IRES-cre sequence was exchanged with the IRES-lacZ-neo unit in the original trap vector by co-electroporation of the vector and pCAGGS-cre into TT2 ES cells containing the trapped Etl4 gene.
