The transgene comprises the human cytomegalovirus (CMV) immediate early gene promoter followed by an SV40 intron, a cDNA encoding N-terminally hemagglutinin- (HA-) tagged mouse MAP kinase-activated protein kinase 5 (MAPKAPK5) in which leucine at amino acid position 337 has been replaced by alanine (L337A), and an SV40 polyadenylation signal. The L337A amino acid substitution confers constitutive activity on the kinase. Expression of the transgene was confirmed by northern blot analysis of RNA isolated from heart tissue. (J:186303)
